I. Product Overview

The program-controlled quantitative sealing machine is an electrically driven device with intelligent circuits and a dedicated internal heating roller, specially designed for modern packaging and production. It is used for sealing 51-well and 97-well quantitative trays, and works with microbial detection reagents to provide a simple, fast and accurate experimental scheme for quantitative detection of total coliforms, fecal (thermotolerant) coliforms and Escherichia coli in water samples.
The 51-well and 97-well quantitative trays are semi-automatic quantitative methods designed based on the traditional Most Probable Number (MPN) statistical model, which can provide quantitative results. This method can replace and outperform traditional methods (membrane filtration and multiple-tube fermentation) for water microbial detection.

For rapid detection, field portable use, emergency monitoring and quantitative detection of total coliforms, fecal (thermotolerant) coliforms and Escherichia coli in water samples.

The enzyme substrate detection system consists of four components: program-controlled quantitative sealing machine, 51/97-well quantitative detection tray, 100 mL quantitative bottle, and enzyme substrate detection reagent.
Applicable water samples: drinking water, source water, bottled water, reclaimed water, secondary water supply, pipe network water, wastewater, food processing water, livestock water, medical water, etc.
Advantages & Features
The enzyme substrate method is a standard method for Escherichia coli detection included in GB 5750-2006. It is currently the most advanced method for coliform detection in water, widely accepted by domestic testing authorities due to its convenience, speed, low false positive rate and suitability for high‑throughput rapid detection.
Compared with multiple-tube fermentation and membrane filtration, the enzyme substrate method greatly simplifies procedures, has low laboratory environment requirements, and shortens detection time to 24 hours. It is highly suitable for routine and emergency monitoring, enabling timely detection and risk prevention.

V. Method Advantages

The machine is used with MMO‑MUG medium (fixed substrate enzyme substrate method) specified in GB 5750‑2006, for sealing water samples in 51‑well or 97‑well quantitative trays.
Working Principle
A water sample containing target bacteria is added to Minimal Medium ONPG‑MUG and incubated at 36℃±1℃.
  • β‑galactosidase from total coliforms hydrolyzes ONPG, turning the medium yellow.

  • β‑glucuronidase from Escherichia coli hydrolyzes MUG, producing characteristic fluorescence.

  • Thermotolerant (fecal) coliforms hydrolyze ONPG at 44.5℃, turning the medium yellow.
